ABSTRACT

Objective To observe the effects of sulforaphane on the apoptosis in human gastric cancer HGC27 cells, and to study the possible mechanism of it. Methods The proliferation activity of HGC27 cells treated with sulforaphane was analyzed by CCK8 kit. Apoptosis rates were assessed by flow cytometry. After HGC27 cells were treated with sulforaphane, and the expression levels of Bcl-2, Bax, caspase-3, PI3K, Akt and p-Akt were measured by Western Blot. After HGC27 cells were treated with LY294002, sulforaphane and LY294002+sulforaphane, the expression levels of PI3K, Akt and p-Akt were also investigated by Western Blot. Results The proliferation of HGC27 cells was significantly suppressed by sulforaphane in dose-dependent and time-dependent manners. The apoptotic rates of HGC27 cells in 10, 20, 40 μg/ml of sulforaphane groups (10.29% ± 1.57%, 23.68% ± 1.69%, 35.29% ± 2.38% vs. 2.52% ± 0.74%) were greatly increased compared with the control group. The expression levels of Bax (18.92 ± 2.18, 34.06 ± 5.06, 44.08 ± 5.69 vs. 12.51 ± 2.15) in 10, 20, 40 μg/ml of sulforaphane groups were greatly increased compared with control group(P<0.05). The expression levels of Bcl-2 (56.39 ± 5.27, 33.06 ± 4.26, 25.61 ± 4.01 vs. 78.25 ± 7.26), PI3K (51.06 ± 5.27, 42.06 ± 5.21, 23.08 ± 4.51 vs. 79.07 ± 8.12), p-Akt/Akt (58.62 ± 5.34, 35.24 ± 4.06, 14.52 ± 2.56 vs. 82.64 ± 8.25) in 10, 20, 40 μg/ml of sulforaphane groups were greatly decreased compared with control group (P<0.05). The expression levels of PI3K (56.41 ± 5.36, 34.37 ± 4.52, 23.11 ± 3.05 vs. 81.24 ± 7.16), p-Akt/Akt (49.52 ± 5.84, 31.06 ± 4.09, 21.05 ± 2.28 vs. 77.52 ± 7.06) in the LY294002, sulforaphane and the LY294002+sulforaphane groups were greatly decreased compared with the control group (P<0.05). Conclusions Sulforaphane may inhibit proliferation and promote apoptosis of HGC27 cells probably by inactivating PI3K/Akt signaling pathway.

ABSTRACT

Aim To investigate the effect of broneol on acute lung injury(ALI) induced by lipopolysaccharide (LPS). Methods Male C57 mice were randomly di-vided into saline group, model group, broneol group and dexamethasone group, then the ALI mouse model was induced by instilling intratracheally with LPS. The levels of tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β),interleukin-6(IL-6) and keratinocyte-de-rived cytokine (KC) were measured at 6h, 12h and 24h after instillation of LPS, and the pathological changes of lung were observed. Mice alveolar macro-phages (MHS) and epithelial cells (MLE-12) were stimulated by LPS. After the stimulation of 1h, 3h, 6h,9h, 12h, 24h, the levels of TNF-α and IL-6 in MHS cells and the contents of KC and macrophage in-flammatory protein-2 (MIP-2) in MLE-12 cells were measured. Results Broneol could inhibit the secre-tion of TNF-α,KC and IL-1β;the early effect of bro-neol on IL-6 was not obvious,but the later effect after the treatment of 24 hours was obvious. After LPS instil-lation 6h and 12h,Broneol could significantly improve lung tissue pathological changes. Broneol had no effect on TNF-α secretion of MHS cells, but it obviously af-fected IL-6 secretion in the later stage. In addition, broneol significantly inhibited KC and MIP2 secretion in MLE-12 cells at the later stage of LPS stimulation. Conclusions Broneol can protect LPS-induced acute lung injury. The mechanism may be related to the inhi-bition of the release of inflammatory factors,the activa-tion of inflammatory cells and the aggregation of neutro-phils.

ABSTRACT

Objective To insight into the status of literature research of drug unpacking in medical institutions in China,to provide decision basis for decision makers to further standardize the management of drugs.Metheds By using the method of systematic review,searching China biomedical literature database (CBM) that articles published before October 15,2016.After two rounds of literature screening,ultimately 67 articles were analyzed.Results The study of drug unpacking in China began in 1997.Of the 67 studies included in this research,a total of 17.9%(12/67) literature was the original study,and a total of 67.2% (45/67) studies were performed in tertiary hospitals.There were 14 studies that reported the objective of drug unpacking in outpatient pharmacy.Some studies reported the manner of drug unpacking,some reported the system construction of drug unpacking in institution,and some reported the common problems of drug unpacking and quality improvement measures.Conclusion At present,there are few original studies on the drug unpacking in China,most of included studies are carried out in tertiary hospitals and focus on the problems because of drug unpacking.The actual size of drug unpacking of the medical institution (the number of varieties,amount) is not known.It is necessary to carry out large-scale field research.

ABSTRACT

Objective To explore the significance of case managers using WeChat platform in the treatment process and extended care for patients with laryngeal cancer.Methods From November 2014 to May 2015, 30 patients with partial resection or total resection of laryngeal cancer in the department of head and neck surgery were selected as a research subjects.Case managers provided fully supports,such as establishing WeChat group,replying online consultations,popularizing health education knowledge,providing psychological counselings.Psychological assessment of the patients was conducted by social support scale(SSRS)and anxiety scale(SAS)in the beginning of case management.Results comparison was performed before and after three months intervention.Results The score of subjective support of patients before intervention was (25.13 ±4.81)points,which after intervention was (28.33 ± 4.41)points.The score of support availability before intervention was (7.20 ±1.99)points,which after intervention was (35.27 ±5.90)points.The differences were statistically significant(t =-2.685,-5.280,2.917,all P <0.05).Conclusion The WeChat platform mediated by the case management is effective in the management of the psychological health of patients with laryngeal cancer,which can effectively promote the rehabilitation of patients.

ABSTRACT

Objective@#To investigate the significance of Twist2 in glioma and whether it is involved in the malignant transformation of glioma by epithelial-mesenchymal transition (EMT).@*Methods@#Using immunohistochemical method detected the expression level of Twist2 in 60 cases of gliomas (including WHO grades Ⅱ, Ⅲ and Ⅳ, each for 20 cases) and 20 cases of non-tumor brain tissues. Real-time fluorescence quantitative PCR and Western blot were used to detect the expression level of Twist2 mRNA and protein in 61 cases of fresh glioma tissue (WHO grade Ⅱ 16 cases, Ⅲ 21 cases, Ⅳ 24 cases) and 12 cases of adjacent tissues, and the expression levels of E-cadherin, N-cadherin and vimentin were also investigated in fresh glioma tissue.@*Results@#Immunohistochemistry results showed that the percentages of Twist2 expression in glioma was 90%(54/60) compared with 30%(6/20) in non-tumor brain tissues(P<0.01). The percentages of Twist2 expression were 75% (15/20), 95% (19/20), and 100% (20/20) in the WHO gradesⅡ, Ⅲ and Ⅳ gliomas, respectively. WHO grades Ⅳ and Ⅲ were significantly higher than that of WHO grade Ⅱ (P<0.01). There was no significant difference between WHO grade Ⅳand WHO Ⅲ glioma (P>0.05). Real-time fluorescence quantitative PCR and Western blot showed that the expression level of Twist 2 in gliomas was significantly higher than that in para-cancerous tissues (P<0.01), and those in WHO grades Ⅳ and Ⅲ gliomas were significantly higher than that in WHO grade Ⅱ glioma (P<0.01). There was no significant difference between WHO grade Ⅳand grade Ⅲ glioma (P>0.05). Detection of key protein expression in EMT by Western blot displayed that the expression of E-cadherin was negatively associated with Twist2 in glioma (r=-0.972, P<0.01). The expression of N-cadherin and vimentin was positively associated with Twist2 in glioma(r=0.971, P<0.01; r=0.968, P<0.01).@*Conclusions@#The expression of Twist2 in human glioma is positively correlated with the malignant grade of glioma, which may be involved in the malignant progression of glioma by EMT.

ABSTRACT

In this work, a simple and fast approach for dipeptide detection was developed. AuNPs were decorated onto the surface of indium tin oxide ( ITO ) glass to serve as the working electrode to trigger the electrochemiluminescence ( ECL) of luminol. The property of this electrode was characterized by transmission electron microscopy ( TEM ) , scanning electron microscopy ( SEM ) , electrochemistry and spectroscopic method. Under the optimum conditions, the dipeptide His-Ala could be detected within a linear range from 2. 44×10-11 mol/L to 1. 22×10-7 mol/L, with a detection limit of 2. 42×10-12 mol/L (S/N=3). In human body, the glucagon-like peptide 1(GLP-1) might lose activity during degradation under the enzymatic action of dipeptidyl peptidase IV (PDD-IV), meanwhile release same concentrated His-Ala dipeptide. Thus, the detection of His-Ala dipeptide was of great significance for investigation of diabetes not only for understanding the relation between GLP-1, PDD-IV and its inhibitor, but also the drug discovering because of its potential availability as the target to control the blood GLP-1 level of type II diabetics.

ABSTRACT

Objective:To investigate the in vitro release behavior and stability of diphenyl diester suspension. Methods:The dis-solution of biphenyl diester dry suspension was detected by HPLC, and the effects of different stirring speed (50, 75, 100 r·min-1 ) and different dissolution media (pH 6. 8 phosphate buffer, 0. 05 mol·L-1 hydrochloric acid solution,water,pH 4. 5 acetate buffer) on dissolution were investigated. The influencing factors testing ( high temperature, high humidity and strong light exposure) , accelerated stability testing[the temperature of (37 ± 5) ℃ and the relative humidity of 76% ± 5%] and long-term stability testing[(25 ± 3) ℃and the relative humidity of 60% ± 5 %] of biphenyl diester dry suspension were carried out. Results:The dissolution behavior of bi-phenyl diester suspension in pH 6. 8 phosphate buffer (50 r·min-1 ) was faster and smoother. The results of influencing factors testing showed that biphenyl diester dry suspension should not be stored under the conditions of high temperature and high humidity. After the samples were stored under the conditions of accelerated testing and long-term stability testing for 6 months, the indicators did not change significantly. Conclusion:The in vitro release of prepared biphenyl diester dry suspension meets the requirements with promis-ing stability.

ABSTRACT

Objective To investigate the CT feature of small cell lung cancer(SCLC),and investigate the clinical value.Methods 55 patients with SCLC and 118 patients with non -small cell lung cancer(NSCLC)were selected as the study subjects.All the patients were confirmed by pathology,the CT feature of lung cancer were com-pared through unenhanced and enhanced scanning by philips brilliance 16 spiral CT.Results There were significant differences between the SCLC group and NSCLC group in tumor type and morphology(χ2 =22.628,21.249,all P <0.01).Central was the mainly type of SCLC,irregular shape and round was the mainly type of NSCLC.The positive rates of positive airway occlusion,pulmonary atelectasis or obstructive pneumonia in central SCLC group and NSCLC group were 57.78% and 86.27%,62.22% and 98.04%,which in central SCLC group were lower than NSCLC group (χ2 =9.830,20.131,all P <0.01).The Hilar and mediastinal lymph nodes positive rate,fusion of lymph node -pos-itive rate,combined positive rate of violations of mediastinal vessels in central SCLC group and NSCLC group were 90.9% and 49.15%,58% and 15.52%,54.55% and 7.63%,which in central SCLC group were greater than the NSCLC group(χ2 =27.887,53.070,47.294,all P <0.01).Conclusion 16 -slice spiral CT can provide important information for diagnosis of SCLC and NSCLC,the CT signs are conducive to the diagnosis of NSCLC,which contai-ning round mass,bronchial smooth,hilar and mediastinal lymph nodes,lymph nodes are not fused with obstructive pneumonia and atelectasis,mediastinal vascular invasion.

Objective:To investigate the incidence trends and clinicopathologic characteristics of papillary thyroid cancer (PTC). Methods:A retrospective analysis was conducting using the following data:3,766 cases with thyroid disease in the People's Hospital of Wuhan University between January 2001 and July 2013;and 977 cases with thyroid cancer in the Hubei Cancer Hospital between Janu-ary 2006 and July 2013. Results:The incidence of thyroid cancer increased significantly since 2008, ranging from 14.94%to 18.10%(P1 cm tumor diameter and multifocal PTC are more likely to be complicated with NLNM.

Objective By using quantum dots-based double-color imaging method to simultaneously label human epidermal growth factor 2 (HER2) on breast cancer cell membrane and the type Ⅳ collagen in the extracellular matrix.To monitor the degree of malignancy of breast cancer and the invasive and metastatic potential.Methods The co-expressions of HER2 and the type Ⅳ collagen in breast cancer of 54 cases were detected and quantified by quantum dots-based double-labeling immunofluorescent histochemistry,and its correlation with clinical pathology parameters and prognosis were analyzed.Results With the increasing HER2 expression level,a progressive decrease in collagen Ⅳ around the cancer nest,the expression of HER2 and the type Ⅳ collagen value was negatively correlated (r =-0.980,P < 0.05) ; the expression of HER2 and collagen Ⅳ was associated with lymph note metastasis,pathological stage (TNM) and disease-free survival(P < 0.05).The expression of HER2 and the type Ⅳ collagen between different age groups,menopause and estrogen receptor (ER) and progesterone receptor (PR) were not significantly different (P > 0.05).Conclusions Quantum dots-based double-color imaging method provides direct observable evidence to support the degradation of HER2 and the type Ⅳ collagen expression,which may help in determining the degree of malignancy and evaluating prognosis.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the clinical significance of atypical squamous cells of unknown significance (ASCUS) with abnormal DNA ploidy in the early diagnosis of cervical lesions.</p><p><b>METHODS</b>Eight thousand four hundred and forty-eight patients were included in this study and all had DNA quantitative analysis and cervical liquid-based cytology. Among 1041 cases with DNA aneuploidy and/or abnormal cervical liquid-based cytology and additional cervical biopsy, histological review was performed in 247 ASCUS cases with abnormal DNA ploidy.</p><p><b>RESULTS</b>(1) Among 8448 cases, 7877 were normal or benign, 426 were ASCUS, 45 were ASC-H, 55 were LSIL and 22 were HSIL by TBS diagnosis. The presence of 1-2 abnormal DNA ploidy cells was detected in 15.3% (65/426) of ASCUS, 11.1% (5/45) of ASC-H, 9.1% (5/55) of LSIL, and 0 (0/22) of HSIL. The presence of ≥ 3 abnormal DNA ploidy cells was detected in 39.0% (166/426) of ASCUS, 75.6% (34/45) of ASC-H, 76.4% (42/55) of LSIL, and 95.5% (21/22) of HSIL. (2) A total of 67 cases of CIN 2, CIN 3 or cancers were found in 247 patients with ASCUS by colposcopy biopsies, of which 13.9% (5/36) had 1-2 abnormal DNA ploidy cells, 45.5% (56/123) had ≥ 3 abnormal DNA ploidy cells and 6.8% (6/88) had normal DNA polidy. ASCUS with 1-2 abnormal DNA ploidy cells and with ≥ 3 abnormal DNA ploidy cells were compared. The difference was statistically significant (χ(2) = 11.79, P < 0.01). But the difference between ASCUS with 1-2 abnormal DNA ploidy cells and normal DNA ploidy had no statistical significance (P > 0.05).</p><p><b>CONCLUSIONS</b>ASCUS with ≥ 3 abnormal DNA ploidy cells has higher risk for developing CIN 2, CIN 3 or invasive carcinoma. The application of DNA quantitative analysis and cervical liquid-based cytology test can help in guiding clinical follow-up and treatment options in patients with ASCUS.</p>

ABSTRACT

Objective To study the role of membrane cytoskeleton linker Ezrin in the growth and metastasis of human breast carcinoma cell line MCF-7.Methods Small interfering RNA(siRNA)targeting Ezrin were transfected into MCF-7 cells.RT-PCR and Western blot were used to detect the down-regulation of Ezrin mRNA and protein.MTT method was used to detect the proliferation of the cells.Flow cytometry was used to detect the cell cycle.Transwell assay was used to detect the migration ability of the transfected MCF-7 cells.Results RT-PCR and Western blot results revealed that Ezrin siRNA down-regulated Ezrin expression at both mRNA and protein levels.Down regulation of Ezrin expression decreased the proliferation rate of the cell line(t=4.479,P<0.01).The cell proportion in G2-M phase (t=-5.997,P<0.01)and the invasion ability(t=5.268,P<0.01)decreased significantly.Conclusion Ezrin plays an important role in the process of proliferation,migration,and invasion of MCF-7 cells.

ABSTRACT

<p><b>OBJECTIVE</b>To evaluate the therapeutic effects of acupotomy-injection with FENG Tian-you spinal manipulation (FSM) for treating cervical spondylotic radiculopathy (CSR).</p><p><b>METHODS</b>Seventy-two patients with CSR were randomly divided into two groups (n = 36 respectively): treatment and control group. The patients in treatment group were treated with acupotomy-injection combined with FSM, and patients in control group were treated with traction. All the patients were followed up for 3 months after treatment and the therapeutic effects were evaluated.</p><p><b>RESULTS</b>Totally there were 72 patients and 69 completed the treatment and the other 3 patients withdraw from treatment. In treatment group, 24 patients were cured, 6 patients had remarkable effects, 4 effective and 2 ineffective; as well in control group, above data were 15, 3, 10 and 8 respectively. The comprehensive effects and the cured patients in the treatment group were obviously better than those of control group (P < 0.05). The comparison of scales for assessment of symptoms and short-form McGill Pain Questionaire before and after treatment in two groups had significant differences (P < 0.001).</p><p><b>CONCLUSION</b>The method of acupotomy-injection combined with FSM for treating CSR is better than conventional traction method.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To evaluate the extensive and degree of physical rehabilitation improvement of the quality of life in laryngectomees.</p><p><b>METHODS</b>Forty nine patients who underwent total laryngectomies were trained by esophageal voice rehabilitation successfully. The questionnaires of performance status scale for head and neck cancer patients (PSS-HN) and the functional assessment of head and neck cancer therapy (FACT-H&N) were answered by them before esophageal voice training and 3 months after successful vocal rehabilitation.</p><p><b>RESULTS</b>Total laryngectomy deteriorated the quality of life in laryngectomees. The mean scores of PSS-HN scale and FACT-H&N questionnaire were lower than the criteria scores after patients underwent total laryngectomy, the mean score were 131. 4,90.6 respectively, the difference was significant statistically (t =53. 673, P <0.001) , (t = 67.44, P <0.001). After successful esophageal speech training, the mean scores of the laryngectomees were improved both in PSS-HN scale and FACT-H&N which were 240.4 and 103.7 respectively, the difference was significant statistically (t = 18.209, P < 0.001) , (t = 21.389, P<0.001).</p><p><b>CONCLUSIONS</b>The quality of life in laryngectomees can be improved by physical rehabilitation and the esophageal voice training.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To present the treatment results and to identify the most effective therapeutic plan of different therapeutic modalities in patients with squamous-cell carcinoma of hypopharynnx.</p><p><b>METHODS</b>A retrospective review of 464 patients with squamous-cell carcinoma of the hypopharynx treated between 1958 and 1998 was accomplished. The clinical characteristics, results of different treatments were analyzed by SPSS 10.0 statistic software.</p><p><b>RESULTS</b>Of 464 patients, the age ranged from 20 to 88 years (mean 56.3 years) and the male to female ratio was 5.5:1. The primary lesion of 383 were originated from the pyriform sinus, 40 from posterior pharyngeal wall and 41 from the postcricoid area. According to the UICC 1997 TNM staging system, 75% had T3 or T4 lesion or 92.2% stage III or IV on presentation. 65% had neck metastases. 202 patients were treated with preoperative radiation plus surgery (R + S), 22 with surgery plus postoperative radiation (S + R), 26 surgery alone (S), 40 patients with salvage surgery after radiotherapy failure (RF) and 174 patients with radiotherapy alone. The overall 5-year survival rate was 34.2%. The overall 5-year survival rate of R + S group was 46.3%, S + R group was 49.2%, S alone group 22.8%, RF group was 40.8%, radiotherapy alone group 18.0% (P < 0.01). The overall 5-year survival rate of R + S group was higher than that of S alone group (P = 0.046). The rate of larynx preservation in R + S group was 39.6% in contrast to that of S + R and S alone group of 16.7% (P = 0.003).</p><p><b>CONCLUSION</b>The survival rate of patients with squamous-cell carcinoma of the hypopharynx treated with combined therapy (R + S or S + R) is better than the other therapeutic modalities. R + S combined is able to offer an obviously higher rate of larynx preservation (39.6% vs 16.7%).</p>

Objective To determine the effective prognostic parameter and the best prognostic index of gastiric carcinoma.Methods The prognostic relevance of clinical and pathological variables were evaluated in 83 patients with histologically proved stomach carcinoma (including the expression of nm23, c erbB2, microbloodvessel and PTNM) by the multivariate analysis of COX regression.Result PTNM staging was the only parameter to enter the COX model.Conclusion The PTNM staging is the most important, reliable and best varible factor in predicting the clinical outcome of stomach cancer.
